About Glyphic:
At Glyphic Biotechnologies, we plan to create the protein revolution for which scientists and researchers have been waiting. We are developing a massively parallel, single-molecule proteome sequencing platform that will transform life science discovery and usher in a new era of insights into human biology and disease. To date, we have raised >$80M from venture partners and non-dilutive grant funding to achieve our vision of next generation proteome sequencing.
What we are looking for in you
We are seeking a Senior or Staff Scientist to join our Assay Development team at a critical stage in platform development. Reporting to the Director of Assay Development, you will be a hands-on expert working at the chemistry-assay interface — developing the chemical methods that enable reliable, high-efficiency sample loading into our expansion assay, and building the novel conjugation, cleavage, and stabilization workflows that power our single-molecule proteome sequencing platform. You will work closely with the Chemistry team to develop sample input methods for new protein and peptide types, including side-chain protection strategies, encoder loading routes, and peptide-from-protein handoff approaches.
The ideal candidate brings deep expertise in peptide chemistry, bioorthogonal and click conjugation strategies, and cleavage chemistry development — with a track record of building new chemical methods for complex, multi-component systems. You combine a synthetic chemist's mechanistic rigor with sensitivity to how chemistry performs inside a demanding multi-cycle assay environment.
This is a full-time, exempt, in-person position with work conducted at our Berkeley, CA location.
What you’ll do
Expansion assay chemistry & sample loading
Optimize bioconjugation methods for peptide loading into the expansion assay; establish robust, high-efficiency loading protocols that perform reliably across diverse peptide populations and physicochemical characteristics.
Identify, characterize, and optimize peptide labeling during the ProSE sequencing assay, leveraging mechanistic understanding of Edman chemistry and amino acid side-chain reactivity and stability.
Develop reagents and methods to maximize the stability of peptides, DNA, and peptide-DNA chimeras throughout the ProSE sequencing assay.
Partner with the Chemistry team to develop amino acid side-chain protection chemistries.
Evaluate compatibility of new chemistries with existing assay components, reagents, and multi-cycle sequencing conditions.
Evaluate emerging chemistries from the literature and internal pipeline; synthesize and test candidate reagents.
Novel assay chemistry workflows
Design and determine the order of operations for chemical manipulations within the assay workflow — including decisions on when specific steps occur on-bead versus off-bead, and how side-chain protection and deprotection strategies are sequenced relative to assay cycles — to ensure compatibility, efficiency, and analytical interpretability.
Optimize selective cleavage chemistry workflows for use within the sequencing assay.
Optimize for efficiency, selectivity, and compatibility with assay conditions.
Develop stabilization strategies for sensitive assay intermediates and conjugates.
Employ analytical methods — HPLC (RP and IEX), LC-MS/MS, UV-Vis, CE — to characterize conjugation efficiency, adduct identity, purity, and stability of assay intermediates and conjugates.
Characterize and troubleshoot the impact of the chemical steps of multi-cycle workflows.
What you need
Required:
Advanced degree in organic chemistry, chemical biology, biochemistry, or a related field
Industry experience designing multi-step chemical workflows in complex biological systems, including sequencing or determining the order of chemical manipulations across assay conditions.
MS/PhD with 6+/4+ years of experience (for Senior Scientist)
MS/PhD with 8+/6+ years of experience (for Staff Scientist)
Demonstrated experience developing site-selective bioconjugation chemistry for native peptides — including N-terminal, C-terminal, and side-chain modification strategies — using click chemistry (CuAAC, SPAAC, IEDDA), NHS-ester coupling, and related bioorthogonal approaches in multi-component peptide systems.
Experience developing multi-step workflows using peptides, oligonucleotides and bioconjugation techniques including in solution and on surfaces.
Proficiency with analytical characterization of complex conjugates and assay intermediates: HPLC (RP and IEX), LC-MS/MS, UV-Vis; demonstrated ability to develop and validate analytical methods for novel chemical entities.
Method-development mindset: history of building and adapting new chemical approaches rather than executing established protocols.
Ability to function as both an independent contributor and a collaborative partner in a cross-functional team environment.
Excellent problem-solving skills and attention to detail.
Excellent written and verbal communication skills, with the ability to work seamlessly with colleagues from diverse scientific and technical backgrounds.

Familiarity with statistical and DOE-based experimental design for chemistry optimization.

What we are looking for in you
Glyphic is seeking a Scientist or Senior Scientist to build and sustain the reagent supply infrastructure that powers our single-molecule proteome sequencing platform. In this role you will own the full reagent lifecycle — from early formulation and characterization through stability qualification, QC method development, and lot release — ensuring that every critical material entering a multi-cycle expansion run meets Glyphic's performance standards.
The ideal candidate brings deep hands-on expertise in biochemical reagent development with a quality-by-design mindset: methodical in experimental approach, rigorous in documentation, and proactive in building the frameworks that sustain consistency at scale. You have experience not just characterizing reagents but qualifying them — setting acceptance criteria, gating go/no-go decisions on incoming lots, and maintaining the provenance records that high-stakes single-molecule runs demand. You are comfortable working at the bench and translating your findings into SOPs, QC protocols, and reagent inventories that the broader Assay Development team can rely on.
This is a full-time, exempt, in-person position with work conducted at our Berkeley, CA location.
What you’ll do
Reagent Development & Formulation
Lead assay reagent development activities, including formulation, characterization, and optimization, for components used in ProSE single-molecule expansion and library preparation workflows, including enzymes, oligos, beads, and solvents.
The materials in scope extend beyond standard biological reagents to include chemically reactive compounds and environmentally sensitive materials susceptible to degradation by oxygen, humidity, light, and temperature fluctuations. Working with these materials demands a chemistry-aware approach to formulation, handling, and storage.
Characterize reagent performance using design-of-experiments (DOE) approaches, systematically varying formulation conditions, buffer composition, excipients, and process parameters to establish operating bounds and define product specifications.
Design and execute experiments to evaluate reagent performance in end-use assay configurations; develop a mechanistic understanding of how reagent composition, lot variability, and environmental sensitivity impact expansion assay outcomes.
Design and execute reagent storage stability studies under accelerated and real-time conditions; develop stability-indicating assays; interpret results against established product specifications and flag out-of-spec trends proactively.
QC Framework & Lot Qualification
Build and implement a QC and control framework for platform reagents, including acceptance criteria, lot release protocols, and QC assay development, to support reliable, consistent supply to the Assay team.
Develop, qualify, and transfer QC test methods for reagent characterization and lot release, including HPLC, fluorescence-based functional assays, and physical characterization methods; set product, process, and incoming material acceptance criteria in collaboration with Assay and cross-functional partners.
Qualify each reagent lot prior to use in multi-cycle or KOL runs; execute hands-on QC testing encompassing physical, chemical, and functional characterization; gate go/no-go decisions on input materials and maintain provenance records for high-stakes runs.
Maintain batch records and quality documentation in accordance with internal standards.
SOPs, Inventory & Continuous Improvement
Establish and maintain SOPs for assay reagents and integrated workflows; systematically investigate lot-to-lot variability and failure modes; troubleshoot deviations and lead root cause analysis and corrective action.
Guard-band critical reagent components and workflow steps to establish the operating bounds of new protocols and define tolerance limits for expansion assay parameters.
Forecast and maintain end-to-end reagent inventory for SOP-level runs, ensuring the Assay team has qualified materials available on schedule for integration runs and platform demonstrations.
Partner with the Automation team to implement and qualify reagent-dependent assay steps on work cell or Hamilton-based platforms; qualify liquid-handling methods against reagent specifications.
Analyze experimental data using statistical methods; synthesize results to identify trends and trace them to upstream reagent, process, or formulation variables; communicate findings through technical reports and presentations, and maintain comprehensive documentation to ensure reproducibility and preserve institutional knowledge.
What you need
Required:
Degree in Biochemistry, Chemical Biology, Biophysics, Biomedical Engineering, or an equivalent field; industry experience including:
PhD / MS / BS with 4+ / 6+ / 8+ years of relevant experience (Senior Scientist)
PhD / MS / BS with 2+ / 4+ / 6+ years of relevant experience (Scientist II)
Proven bench expertise in reagent development or analytical science, with hands-on experience across biochemical characterization techniques with the ability to establish operating bounds and guard bands for critical reagent and process parameters.
Deep understanding of reagent formulation principles and their influence on stability and functional performance; with practical experience across the full reagent development cycle including formulation, stability studies, and QC method development.
Experience working with chemically reactive or environmentally sensitive compounds, including materials susceptible to degradation by oxygen, humidity, or light, with appropriate handling, storage, and stability assessment practices
Strong experimental design skills, with the ability to plan and execute DOE-informed studies and interpret results against specifications.
Experience developing reagents for multistep, integrated assay workflows involving multiple handoff points or checkpoints, including multi-omics or multi-component platforms.
Experience establishing and maintaining robust SOPs for end-to-end integrated workflows, with consistent capture of key experimental variables, reagents, and process conditions — enabling cross-functional traceability and correlation with downstream data and performance.
Excellent problem-solving skills and meticulous attention to detail.
Strong written and verbal communication skills, with the ability to work effectively with colleagues across diverse scientific and technical backgrounds.
Comfortable navigating a fast-paced, resource-constrained environment with shifting priorities.
Nice to have:
Background in proteomics reagents, protein modification chemistry, peptide-oligonucleotide conjugates, or single-molecule assay components.
Experience with automation platforms for liquid handling (e.g., Hamilton, ThermoFisher, Agilent) and protocol validation on automated workcells.

Estimated Base Salary $111,725 - $161,900
This is the pay range for this position that we reasonably expect to pay. Individual compensation is based on various factors including, experience, education, skillset, and geographic location. This range is for the SF Bay Area, California location and may be adjusted to the labor market in other geographic areas.

What we are looking for in you
We are seeking a Senior or Staff Scientist, Bioconjugation to join our Chemistry team at a pivotal stage in assay development. Reporting to the Senior Director of Chemistry, you will be a hands-on expert who designs, develops, and executes bioconjugation strategies for peptide and oligonucleotide components that are central to our single-molecule proteome sequencing assay.
The ideal candidate brings deep expertise in protein and peptide bioconjugation, site-specific chemical modification of amino acid sidechains, and oligonucleotide-biomolecule conjugation, with a track record of working in complex, multi-component systems (e.g., ADCs, therapeutic bioconjugates, or new bioconjugated components for assays). A successful candidate will have an innovative method-development mindset and will effectively build new approaches to making bioconjugates. This individual will thrive in a cross-functional environment where chemical solutions must be tightly integrated into assay design.
This role offers the unique opportunity to develop first-in-class conjugation chemistry that enables single-molecule protein sequencing.
This is a full-time, exempt, in-person position with work conducted at our Berkeley, CA location.
What you’ll do
Bioconjugation strategies:
Partner with sample prep and assay development to screen and select conjugation chemistries for peptides and oligonucleotides that enable single molecule protein sequencing
Design bioconjugation workflows using NHS-ester chemistry, maleimides, isothiocyanate chemistry, and click chemistry (CuAAC, SPAAC, IEDDA)
Prepare bioconjugation reagents to enable studies from proof-of-concept through prototype-kitting.
Peptide chemistry:
Develop labeling strategies based on Edman Chemistry
Develop and apply selective C-terminal and N-terminal conjugation chemistries
Develop site-specific labeling strategies for amino acid sidechains (Cys, Lys, Arg, Tyr, Trp, His, Asp/Glu) on native peptides. Screen protecting groups to enhance amino acid signal in the protein sequencing assay
Enable on-bead workflow to label and release peptides for interrogation in the sequencing assay
Synthesize, characterize, and scale:
Synthesize and purify peptide and oligonucleotide conjugates for assay testing
Characterize conjugation efficiency, purity, and stability using tools such as HPLC, LC-MS/MS, NMR, CE, UV/Vis, and FTIR
Scale conjugate synthesis from R&D quantities to prototype-grade supply for internal testing, with attention to reproducibility, shelf-life, and fit-for-purpose quality
Collaborate and document:
Work cross-functionally with sample preparation, assay integration, and reagent manufacturing teams to troubleshoot conjugation failures and optimize workflows
Contribute to intellectual property development through invention disclosures
Maintain rigorous experimental documentation in Confluence and laboratory notebooks; author and maintain SOPs for new conjugation methods
What you need
Required:
MS or PhD in organic chemistry, chemical biology, biochemistry, or a related field
Industry experience in bioconjugation chemistry, with demonstrated ability to independently design, develop, and troubleshoot conjugation workflows
MS/PhD with 8+/6+ years of experience (for Staff Scientist)
MS/PhD with 6+/4+ years of experience (for Senior Scientist)
Deep expertise in protein and/or peptide bioconjugation: NHS esters, maleimides, click chemistries (CuAAC, SPAAC, IEDDA), isothiocyanate coupling, and related bioorthogonal approaches
Experience with site-specific amino acid sidechain chemistry beyond maleimide chemistry (e.g. Cys, Lys, Arg, Tyr, Trp, Asp/Glu) for application in multistep workflows
Track record developing novel blocking groups and conducting SAR studies to optimize blocking group performance and selectivity
Proficiency in analytical characterization: HPLC method development, LC-MS/MS for conjugate analysis, UV/Vis, and CE
Method-development mindset: history of building and adapting new chemical approaches rather than executing established protocols
Demonstrated ability to function as an independent contributor and collaborative partner in a cross-functional team setting
Nice to have:
Hands-on experience with solid-phase peptide synthesis (SPPS), including protecting group strategies beyond standard Fmoc/Boc and selective on-resin modifications
Experience with peptide C-terminal chemistry manipulation methods: thioester formation, native chemical ligation, or photoredox/iridium-catalyzed C-terminal labeling
Familiarity with oligonucleotide chemistry and oligo-conjugation techniques (ITC, phosphoramidite modifications, click coupling to nucleic acids)
Experience with scalable surface chemistry methods for flow cells or solid-phase supports
Familiarity with statistical and DOE-based experimental design for chemistry optimization
Prior experience in an early-stage biotech or tools/diagnostics company

- Optimize performance of large-scale ML systems, including distributed training, inference efficiency, and GPU-level optimizations where necessary.
